The End of Botox Animal Tests Draws Closer: Europe Moves to Scrap the Mouse Test

To this day, mice suffocate for smooth foreheads. That may soon end: the European Directorate for the Quality of Medicines (EDQM) has announced it will gradually remove the cruel mouse test for Botox from the European Pharmacopoeia. Doctors Against Animal Experiments (Ärzte gegen Tierversuche) calls it the payoff of almost two decades of campaigning.

The test in question

Botulinum toxin, better known as Botox, has medical uses, for instance against eyelid spasms or certain movement disorders. Mostly, though, the nerve toxin serves to smooth wrinkles. For quality control, mice are exposed to varying doses: they suffer paralysis and respiratory distress, and many suffocate in agony. The so-called LD50 test determines the dose lethal to half of the animals and is classified as “severe” under EU law, the highest severity grade.

The absurd part: reliable cell-based, animal-free test methods have been available for years. How many mice are used annually for Botox tests, for which purposes and by which companies, remains untransparent to this day, another point ÄgT criticises. Almost two decades of pressure

Ärzte gegen Tierversuche and the European Coalition to End Animal Experiments (ECEAE) have fought the Botox tests for almost twenty years. In 2023 they handed more than 164,000 signatures from across Europe to the European Medicines Agency (EMA), which then put the issue on its agenda. The campaigns “Stoppt Botox-Tierversuche” (ÄgT) and “Stop Botox Animal Testing!” (ECEAE) are currently running.

First the rabbits, now the mice

The EDQM's announcement carries a remarkable title: “First rabbits, now mice”. It describes a pattern: the European Pharmacopoeia has already scrapped the pyrogen test on rabbits (article in German), and now the Botox mouse test is next. Animal-free methods are replacing, one by one, tests that were considered irreplaceable for decades.
